Description: The Ras superfamily of small guanosine triphosphatases (GTPases) is traditionally subdivided into five families: Ras, Rho, Rab, Ran, and Arf. Rap1/Krev1 is a member of the Ras family of low molecular weight GTP-binding proteins1. Ras-like GTPases are ubiquitously expressed, evolutionarily conserved molecular switches that couple extracellular signals to various cellular responses. Rap1 is primarily found at the cytosolic side of intracellular membranes and has two isoforms: Rap1a and 1b2. Both isoforms have a molecular mass of 21 kDa and are isoprenylated at the carboxyl-terminal and phosphorylated by the cAMP-dependent protein kinase A (PKA)3. Rap1 cycles between a GTP-bound active form and a GDP-bound inactive form that is mediated by GTPase activating protein (GAP) and GDP dissociation stimulator (GDS)4,5. Activation occurs by a variety of extracellular stimuli through several conserved guanine nucleotide exchange factors (GEFs) and GTPase activating proteins (GAPs). Rap1 is proposed to regulate Ras mediated signaling and may also be involved in the regulation of integrinmediated cell adhesion, although the mechanism of regulation is not known. Overexpression of Rap reverses the transformed phenotype induced by Ras, possibly by competing with Ras for interaction with Ras-GAP. Rap has been shown to participate in MAP kinase cascade activated by growth factor and maintaining human T cell anergic state by blocking IL-2 expression.

Description: Glycogen Synthase Kinase 3β (GSK-3β) is a unique serine/threonine kinase that is inactivated by phosphorylation. In response to insulin binding, PKB/AKT phosphorylates GSK-3β on serine 9, which prevents GSK-3β from phosphorylating glycogen synthase. Unphosphorylated glycogen synthase is active and able to synthesize glycogen. GSK-3β is also unique in that it requires a substrate that has been phosphorylated by a distinct kinase before it can phosphorylate the substrate. This phosphate priming mechanism explains why phosphorylation of serine 9 inactivates GSK-3β. The phosphorylated serine binds to the GSK-3β priming phosphate position and prevents binding of alternative substrates. In addition to insulin signaling, GSK-3β participates in the Wnt signaling pathway, where it forms a complex with axin, beta-catenin and adenomatous polyposis coli (APC) protein. In the presence of Wnts, GSK-3β is unable to phosphorylate beta-catenin, which leads to stabilization of beta-catenin. The Wnt pathway inactivates GSK-3β via the proteins, Dishevelled and FRAT, which disrupt the interaction of GSK-3β with axin, beta-catenin, and APC. Clinically, there is considerable interest in GSK-3β inhibitors because they may mimic the effect of insulin or reduce the hyperphosphorylation of Tau that is observed in Alzheimer's Disease.

Description: The diffusible free radical gas nitric oxide (NO) affects a variety of physiological functions, and is a key regulator of the cardiovascular, nervous, and immune systems. NO is synthesized in many tissues from L-arginine, oxygen, and NADPH by three known isoforms of a heme-containing flavoprotein termed NO synthase (nNOS/NOS-I, iNOS/NOS-II, and eNOS/NOS-III). nNOS is a constitutively expressed neuronal NOS isoform that exits in its latent form until it is activated by the binding of calmodulin follo wing elevation of intracellular calcium levels. The C-terminus of nNOS contains a conserved serine residue, Ser1417, analogous to Ser1177 of the constitutively expressed endothelial NOS isoform (eNOS). Phosphorylation of Ser1417 is believed to regulate nNOS activation, particularly in glucose-sensing neurons, where inhibition of AMPK pathways by glucose and leptin serve to suppress nNOS activity, whereas activation of AMPK by insulin leads to nNOS activation.
