Description: PAG [Cbp] is a ubiquitously expressed transmembrane adapter protein. Depending on its phosphorylation status, it appears as diffuse band(s) of 80-90kDa by SDS-PAGE, with phosphorylation leading to a shift towards a higher apparent molecular mass. Tyrosine phosphorylated PAG recruits Csk to the membrane and negatively regulates Src family kinases via Csk-mediated phosphorylation. Upon T cell receptor engagement, PAG becomes dephosphorylated, thereby displacing Csk from the membrane and enabling the activation of the Src kinases Fyn and Lck. PAG negatively regulates TCR (T cell antigen receptor)-mediated signaling in T cells and FCER1 (high affinity immunoglobulin epsilon receptor)-mediated signaling in mast cells. It promotes CSK activation and recruitment to lipid rafts, which results in LCK inhibition. It inhibits immunological synapse formation by preventing dynamic arrangement of lipid raft proteins and may be involved in cell adhesion signaling.

Description: ORAI3 Monoclonal Antibody: Antigen stimulation of immune cells triggers Ca++ entry t hrough Ca++ release-activated Ca++ (CRAC) channels. ORAI3 is one of two mammalian homologs to ORAI1, a recently identified four-transmembrane spanning protein that is an essential component of CRAC. All three homologs have been shown to function as Ca++ plasma membrane channels gated through interactions with STIM1, the store-activated endoplasmic reticulum Ca++ sensor. However, ORAI3 channels failed to produce detectable Ca++ selective currents in cells co-transfected with ORAI3 and STIM1, indicating that ORAI3 channels undergo a lesser degree of depotentiation than ORAI1 or ORAI2. Na+ currents through ORAI1, 2 and 3 channels were equally inhibited by extracellular Ca++, indicating that each have similar affinities for Ca++ within the selectivity filter. This antibody is predicted to have no cross-reactivity to ORAI1 or ORAI2. Larger molecular weight bands are sometimes seen in SDS-PAGE; these may represent post-translationally modified ORAI 3.

Description: Collagens are highly conserved throughout evolution and are characterized by an uninterrupted "Glycine-X-Y" triplet repeat that is a necessary part of the triple helical structure. For these reasons, it is often extremely difficult to generate antibodies with specificities to collagens. The development of type specific antibodies is dependent on NON-DENATURED three-dimensional epitopes. Collagens are extensively purified for immunization from human and bovine placenta and cartilage by limited pepsin digestion and selective salt precipitation. This preparation results in a native conformation of the protein. Antibodies are isolated from rabbit antiserum and are extensively cross-adsorbed by immunoaffinity purification to produce 'type' specific antibodies. Greatly diminished reactivity and selectivity of these antibodies will result if denaturing and reducing conditions are used for SDS-PAGE and immunoblotting. This antibody reacts with most mammalian Type IV collagens and has negligible cross-reactivity with type I, II, III, V, or VI collagens.

Description: Pst1 Antibody: Protoplast secreted protein 1 (Pst1) is an anchored plasma membrane protein. Pst1 was previously identified as a protein secreted by yeast regenerating protoplasts, which suggests a role in cell wall construction. Pst1 is a protein with 444 amino acids and is attached to yeast cell wall via a glycosylphosphatidylinisotol (GPI) anchor. Pst1 contains 15 potential N-linked glycosylation sites and is heavily glycosylated. It migrates at approximately 200 kDa on SDS-PAGE when produced in wild type S. cerevisiae. Our Pst1 protein (Catalog No. 95-151) was expressed in a genetically manipulated triple-mutant (TM) S. cerevisiae stain ( Delta och1 Delta mnn1 Delta mnn4), which results in the production of sole Man8GlcNAc2 carbohydrate structures and Pst1 migration at approximately 100 kDa. When produced in the TM yeast, Pst1 can be recognized by several glycan-specific HIV-1 broadly neutralizing antibodies, including 2G12 and recently identified PGT antibodies. Among several heavily N-glycosylated TM yeast glycoproteins, Pst1 shows higher affinity for 2G12 and efficiently inhibits gp120 interactions with 2G12 and DC-SIGN, and it also blocks 2G12-mediated neutralization of HIV-1 pseudoviruses.

Description: Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide-stimulated macrophages, and causes tumor necrosis when injected into tumor bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia. TNF alpha exists as a multimer of two, three, or five non-covalently linked units, but shows a single 17 kDa band following SDS PAGE under non-reducing conditions. TNF alpha is closely related to the 25 kDa protein Tumor Necrosis Factor beta (lymphotoxin), sharing the same receptors and cellular actions. TNF alpha causes cytolysis of certain transformed cells, being synergistic with interferon gamma in its cytotoxicity. Although it has little effect on many cultured normal human cells, TNF alpha appears to be directly toxic to vascular endothelial cells. Other actions of TNF alpha include stimulating growth of human fibroblasts and other cell lines, activating polymorphonuclear neutrophils and osteoclasts, and induction of interleukin 1, prostaglandin E2 and collagenase production. TNF alpha is currently being evaluated in treatment of certain cancers and AIDS Related Complex.

Description: Microtubules are 25nm diameter protein rods found in most kinds of eukaryotic cells. They are polymerized from a dimeric subunit made of one a subunit and one b tubulin subunit. Microtubules are associated with a family of proteins called microtubule associated proteins (MAPs), which includes the protein τ and a group of proteins referred to as MAP1, MAP2, MAP3, MAP4 and MAP5 (Kindler & Gardner 1994). MAP2 is made up of two ~280 kDa apparent molecular weight bands referred to as MAP2a and MAP2b. A third lower molecular weight form, usually called MAP2c, corresponds to a pair of protein bands running at ~70 kDa on SDS-PAGE gels. All these MAP2 forms are derived from a single gene by alternate transcription, and all share a C-terminal sequence which includes either three or four microtubule binding peptide sequences, which are very similar to those found in the related microtubule binding protein τ. MAP2 isoforms are expressed only in neuronal cells and specifically in the perikarya and dendrites of these cells. MAP2 has been recently shown to be the specific receptor for the neurosteroid pregnenolone (Fontaine-Lenore V. et al., 2006).
