Description: DIP2A, also known as DIP2, is a 1571 amino acid nuclear protein. It is one of three human homologs (DIP2A, DIP2B and DIP2C) of the Drosophila dip2 (disconnected-interacting protein 2) protein. In Drosophila, dip2 interacts with disco, a protein required for neuronal connections in the visual systems of larvae and adults. The closest vertebrate homologs to disco are the basonuclin genes. In mice, DIP2 homologs show restricted expression to the brain. This suggests that, similar to the function of Drosophila dip2, vertebrate DIP2 homologs may play a role in the development of the nervous system. Expressed ubiquitously with highest expression in the brain, DIP2A is thought to function in signaling throughout the central nervous system by providing positional clues for axon patterning and pathfinding. Four isoforms of DIP2A exist due to alternative splicing events.

LRRTM4 (leucine-rich repeat transmembrane neuronal protein 4) is a 590 amino acid member of the LRRTM protein family. Expressed in neuronal tissues, LRRTM4 may play a role in the development and maintenance of the vertebrate nervous system. A single-pass type I membrane protein, LRRTM4 contains 10 LRR repeats. LRRTM4 is expressed as two isoforms produced by alternative splicing.

Description: HERPUD1 is a includes the inhibition of translation to prevent further accumulation of unfolded proteins, the increased expression of proteins involved in polypeptide folding, known as the unfolded protein response (UPR), and the destruction of misfolded proteins by the ER-associated protein degradation (ERAD) system. This gene may play a role in both UPR and ERAD. Its expression is induced by UPR and it has an ER stress response element in its promoter region while the encoded protein has an N-terminal ubiquitin-like domain which may interact with the ERAD system. This protein has been shown to interact with presenilin proteins and to increase the level of amyloid-beta protein following its overexpression. Alternative splicing of this gene produces multiple transcript variants encoding different isoforms. The full-length nature of all transcript variants has not been determined.

Robust expression in neural systems associated with cognitive, motor and sensory functions is consistent with the myriad of symptoms experienced by patients with triple A syndrome.
