Description: Transforming Growth Factor beta 5 (TGF beta 5) is a member of the TGF beta family of growth factors. The TGF beta polypeptides are multifunctional; capable of influencing cell proliferation, differentiation, and other functions in a wide range of cell types. Transformed, as well as nonneoplastic tissues, release transforming growth factors; and essentially all mammalian cells possess a specific TGF receptor. The multi-modal nature of TGF beta is seen in its ability to stimulate or inhibit cellular proliferation. In general, cells of mesenchymal origin appear to be stimulated by TGF beta whereas cells of epithelial or neuroectodermal origin are inhibited by the peptide.

Description: Induces apoptosis in tumorigenic and transformed cell lines, involved in T cell mediated cytotoxicity, TRAIL ligand of cognate receptors induces NF-kappa-B signaling in a variety of cell types. TRAIL is a 30kDa protein known as TNF-related apoptosis inducing ligand, CD253, TNFSF10, or Apo-2L. It can be expressed as a cell surface (30kDa) and soluble ligand (19-20kDa) produced by megakaryocytes and platelets, monocytes, neutrophils, NK cells and activated T cells. This antigen is also expressed on tumor cells and transformed cell lines. TRAIL has been reported to induce apoptosis in tumor and transformed cell lines by a caspase-dependent process. This antibody is useful for flow cytometric staining and blocking studies.

Description: When DNA is subcloned in the pUC polylinker region, β-galactosidase production is interrupted, resulting in the inability of cells to hydrolyse X-Gal. This results in the production of white colonies amongst a background of blue colonies. This experiment provides a DNA fragment together with a linear plasmid and T4 DNA ligase. Following the ligation to synthesis the recombinant plasmid, competent <i>E. coli</i> cells are transformed and the number of recombinant antibiotic-resistant white and blue colonies are counted. β-galactosidase activity is assayed from blue and white bacterial cells.

Description: The early region (E1) of the adenovirus genome, responsible for transforming activity, is localized within the left most 11% of the viral genome and consists of two transcriptional units E1A and E1B. E1A is sufficient for partial transformation and immortalization of primary cells. E1A gene products are necessary for normal levels of transcription of the other early regions of the adenovirus genome during productive infection and are able to either activate or repress the transcription of specific cellular genes. E1A forms specific complexes with cellular proteins including p105 causing inhibition of the cell cycle inducing arresting function of p105.

Description: Minimum Essential Medium (MEM) with Earle’s Balanced Salts is a modification of Eagle’s earlier Basal Medium (BME) which contains a higher concentration of essential nutrients. These media promote the growth of a variety of normal and transformed cells. Since they contain Earle’s Balanced Salts, they are suitable for use in atmospheres charged with CO₂ gas.

Description: SKI is a nuclear protein that forms homodimers and heterodimers and binds to DNA to function as transcriptional activators and repressors. The SKI oncoprotein dramatically affects cell growth, differentiation, and/or survival. SKI was shown to act in distinct signaling pathways including those involving nuclear receptors, transforming growth factor beta, and tumor suppressors. SKI inhibits transforming growth factor-beta (TGF-beta) signaling through interaction with Smad proteins. SKI represses Smad-mediated transcriptional activation, probably through its action as a transcriptional co-repressor. SKI also inhibits TGF-beta-induced downregulation of genes such as c-myc.

Description: Transcriptional transactivator. Binds double-stranded DNA (By similarity). Has transforming activity. Inactivates, with E6-AP ubiquitin-protein ligase, the human p53/TP53 tumor suppressor protein by targeting it to degradation. Binds and targets human MUPP1/MPDZ protein to degradation. Those two functions presumably contribute to transforming activity. Interaction with human FBLN1 protein also seems to be linked to cell transformation.
