Description: Recognizes a protein of ~76 kDa, which is identified as Nucleolin (NCL). It is the major nucleolar phosphoprotein of growing eukaryotic cells. NCL is located mainly in dense fibrillar regions of the nucleolus. It is found associated with intranucleolar chromatin and pre-ribosomal particles. Human NCL gene consists of 14 exons with 13 introns and spans approximately 11kb. It induces chromatin decondensation by binding to histone H1. It is thought to play a role in pre-rRNA transcription and ribosome assembly.This MAb can be used to stain the nucleoli in cell or tissue preparations and can be used as a marker of the nucleoli in subcellular fractions. It produces a speckled pattern in the nuclei of cells of normal and malignant cells and may be used to stain the nucleoli of cells in fixed or frozen tissue sections. It can be used with paraformaldehyde fixed frozen tissue or cell preparations and formalin fixed, paraffin-embedded tissue sections.

Description: Laminins are large hetero-trimeric, non-collagenous glycoproteins composed of α, β, and γ chains. This MAb reacts with laminin B2/1 chain of ~210 kDa and does not cross-react with other basement membrane components or fibronectin. Its specificity was established by immunoprecipitation and immunofluorescence of human skeletal muscle and kidney with laminin chain-specific MAbs. Epithelial sheets in vivo are separated from the mesenchymal elements of the stroma by a thin layer of a specialized type of extracellular matrix termed the basement membrane (BM). This structure consists of individual components, some of which are ubiquitous in BMs and some are not. The ubiquitous ones comprise laminin (LN), entactin/nidogen (EN), collagen type IV (CIV), and large heparan sulfate proteoglycan (HSPG), which interact specifically with each other to form a continuous and regular BM. Alterations of BM integrity, from local discontinuities up to complete loss, are described in many types of human and animal epithelial neoplasms. This MAb stains uniformly all human and murine basement membranes.

Description: Recognizes a 53 kDa protein, identified as the Ornithine Decarboxylase (ODC-1). ODC is the initial and rate-limiting enzyme in the biosynthetic pathway of polyamines and is involved in the conversion of ornithine to putrescine. The biological activity of ODC-1 is rapidly induced in response to virtually all agents known to promote cell proliferation including hormones, drugs, growth factors, mitogens, and tumor promoters. Reportedly, ODC mRNA levels are elevated in lung carcinomas as well as in colon adenomas and carcinomas. ODC activity in colorectal carcinomas is greater than those in adenomas and normal mucosa.

MAb HSA98 binds to human hepatocytes and the majority of human hepatocellular carcinomas (HCC's). In frozen sections, it stains hepatic cells and may be used as a marker of the liver. Cell preparations of hepatocellular carcinoma biopsies or cell lines are found to bind HSA98 on the cell surface. This MAb stains liver hepatocytes in frozen human liver sections and is positive on the cell surface of human liver carcinomas.

Description: Membrane-permeant form of Rhod-2 that can be loaded into cells via incubation. Because of the relatively low water solubility of the AM ester, the mild detergent Pluronic F-127 is often used as a dispersing agent to facilitate dye loading. It is found on a wide variety of tumor cells including follicular and papillary carcinoma of thyroid, adenocarcinomas from endometrium, lung, ovary, pancreas, and breast as well as malignant melanoma, endodermal sinus tumor, and small cell carcinoma.
