Description: Recognizes a cell surface protein of 220-240 kDa, identified as L1 cell adhesion molecule. The L1CAM gene, which is located in Xq28, is involved in three distinct conditions: 1) HSAS (hydrocephalus-stenosis of the aqueduct of Sylvius); 2) MASA (mental retardation, aphasia, shuffling gait, and adducted thumbs); and 3) SPG1 (spastic paraplegia). The L1, neural cell adhesion molecule (L1CAM) also plays an important role in axon growth, fasciculation, and neural migration as well as in mediating neuronal differentiation. Expression of L1 protein is restricted to tissues arising from neuroectoderm. This MAb is useful in the identification of primitive neuroectodermal tumors. It binds to tumors of neuroectodermal and glial origin e.g. neuroblastoma and Schwannomas. It does not bind to pediatric or adult brain.

Description: The mouse monoclonal antibody recognizes CD1b, a 44 kDa type I glycoprotein associated with beta2-microglobulin. It is expressed on dendritic cells, Langerhans cells, thymocytes, and T acute lymphoblastic leukemia cells. The CD1 multigene family encodes five forms of the CD1 T-cell surface glycoprotein in human, designated CD1A, 1B, 1C, 1D and 1E. CD1, a type 1 membrane protein, has structural similarity to the MHC class I antigen and has been shown to present lipid antigens for recognition by T lymphocytes. Constitutive endocytosis of CD1B molecules and the differential sorting of MHC class II from lysosomes separate peptide- and lipid antigen-presenting molecules during dendritic cell maturation. CD1B is also expressed in interdigitating cells.

Description: Recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. IgM antibody is prominent in early immune responses to most antigens. Aberrant levels are associated with immune deficiency states, hereditary deficiencies, myeloma, Waldenstrom's macroglobulinemia, chronic infection and hepatocellular disease. This MAb is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The most common feature of these malignancies is the restricted expression of a single heavy chain class. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.

Description: At least five CD1 genes (CD1a, b, c, d, and e) are identified. CD1 proteins have been demonstrated to restrict T cell response to non-peptide lipid and glycolipid antigens and play a role in non-classical antigen presentation. CD1a is a non-polymorphic MHC Class 1 related cell surface glycoprotein, expressed in association with Beta-2 microglobulin. Anti-CD1a labels Langerhans cell histiocytosis (Histiocytosis X), extranodal histiocytic sarcoma, a subset of T-lymphoblastic lymphoma/leukemia, and interdigitating dendritic cell sarcoma of the lymph node. When combined with antibodies against TTF-1 and CD5, anti-CD1a is useful in distinguishing between pulmonary and thymic neoplasms since CD1a is consistently expressed in thymic lymphocytes in both typical and atypical thymomas, but only focally in 1/6 of thymic carcinomas and not in lymphocytes in pulmonary neoplasms. Anti-CD1a is reported to be a new marker for perivascular epithelial cell tumor (PEComa).

Description: Recognizes an oncofetal antigen of 220 kDa, identified as a tumor-associated glycoprotein (TAG-72) with properties of a mucin. This MAb defines the mucin-carried sialylated-Tn epitope. TAG-72 is usually expressed by adenocarcinomas, but is negative in mesotheliomas. Studies have reported that this antibody has 80% sensitivity and 93% specificity for pulmonary adenocarcinoma. Therefore, TAG-72 is a useful marker to distinguish between mesothelioma and adenocarcinoma. However, false positive reactions can occur so results must be interpreted with the utmost caution. This antibody may be useful in the differentiation of non-small cell carcinomas from small cell carcinomas of the lung. The combined use of anti-TAG-72 and anti-GCDFP-15 is valuable in the diagnosis of apocrine carcinoma.

Description: MAb 2H11 reacts with a partially defined epitope of human thyroglobulin. This epitope is different form the epitope recognized by MAb 6E1. Thyroglobulin is a 660 kDa dimeric pre-protein with mutiple glycosylation sites. It is produced by and processed within the thyroid gland to produce the hormone thyroxine and triiodothyronine. Prior to forming dimers, thyroglobulin monomers undergo conformational maturation in the endoplasmic reticulation. The vast majority of follicular carcinomas of the thyroid will give positive immunoreactivity for anti-thyroglobulin even though sometimes only focally. Poorly differentiated carcinomas of the thyroid are frequently anti-thyroglobulin negative. Adenocarcinomas of other-than-thyroid origin do not react with this antibody. This antibody is useful in identification of thyroid carcinoma of the papillary and follicular types. Presence of thyroglobulin in metastatic lesions establishes the thyroid origin of tumor. Anti-thyroglobulin, combined with anti-calcitonin, can identify medullary carcinomas of the thyroid. Furthermore, anti-thyroglobulin, combined with anti-TTF1, can be a reliable marker to differentiate between primary thyroid and lung neoplasms.

Description: Recognizes a protein of 110 kDa, which is identified as androgen receptor (AR). It reacts with full length, and the newly described A form of the receptor. It does not cross react with estrogen, progesterone, or glucocorticoid receptors. The expression of AR is reportedly inversely correlated with histologic grade i.e. well differentiated prostate tumors show higher expression than the poorly differentiated tumors. In prostate cancer, AR has been proposed, as a marker of hormone-responsiveness and thus it may be useful in identifying patients likely to benefit from anti-androgen therapy. Anti-androgen receptor has been useful clinically in differentiating morpheaform basal cell carcinoma (mBCC) from desmoplastic trichoepithelioma (DTE) in the skin.This MAb is superb for staining of formalin/paraffin tissues.

Description: This MAb recognizes an antigen associated with the Golgi complex in human cells only. It can be used to stain the Golgi complex in cell or tissue preparations and can be used as a Golgi marker in subcellular fractions. It produces a diffuse staining pattern of the Golgi zone in normal and malignant cells. This MAb is an excellent marker for human cells in xenographic model research. It reacts specifically with human cells. The Golgi apparatus is an organelle present in all eukaryotic cells that forms a part of the endomembrane system. The primary function of the Golgi apparatus is to process and package macromolecules synthesized by the cell for exocytosis or use within the cell. The Golgi is made up of a stack of flattened, membrane-bound sacs known as cisternae, with three functional regions: the cis face, medial region and trans face. Each region consists of various enzymes that selectively modify the macromolecules passing though them, depending on where they are destined to reside. Several spherical vesicles that have budded off of the Golgi are present surrounding the main cisternae.

GFAP is specifically found in astroglia. GFAP is a very popular marker for localizing benign astrocyte and neoplastic cells of glial origin in the central nervous system. Antibody to GFAP is useful in differentiating primary gliomas from metastatic lesions in the brain and for documenting astrocytic differentiation in tumors outside the CNS.

The CC chemokine receptor 5 (CCR5) is a member of the CC-chemokine receptor family, and has the characteristic structure of a 7 transmembrane G protein-coupled receptor (GPCR). CCR5 regulates trafficking and effector functions of memory/effector Th1 cells, macrophages, NK cells, and immature dendritic cells. CCR5 and its ligands play an important role in viral pathogenesis. CCR5 represents the co-receptor for macrophage (M) and dual (T cell and M)-tropic immunodeficiency viruses. Together with the CD4 binding receptor, CCR5 plays a critical role in HIV entry into the target cells. Moreover, the CCR5 ligands macrophage inflammatory protein (MIP)-1 alpha, MIP-1 beta and RANTES act as endogenous inhibitors of HIV infection, making both CCR5 and its chemokine ligands attractive therapeutic targets for HIV infection. Recent studies have also highlighted the role of CCR5 in a variety of other human diseases, ranging from infectious and inflammatory diseases to cancer.

Description: Recognizes a 14 kDa protein, identified as S100A9 (also known as Calgranulin B or MRP-14); expressed by granulocytes, monocytes and by tissue macrophages.The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. Altered expression of this protein is associated with the disease cystic fibrosis. This MAb reacts with neutrophils, monocytes and macrophages, and has been shown as an important marker for identifying macrophages in tissue sections. Among cells that are now recognized as macrophages are histiocytes, Kupffer cells, osteoclasts, microglial cells, synovial type A cells, interdigitating cells, and Langerhans cells (in normal tissues) and epithelioid cells and Langerhans-type and foreign-body-type multinucleated giant cells (in inflamed tissues).

It also stains the mantle zone B-lymphocytes of the lymph node and spleen, spermatogonia, and chief cells of the stomach. S100A9 is expressed by macrophages in acutely inflamed tissues and in chronic inflammation. It is detected in peripheral blood leukocytes, in neutrophils and granulocytes. It is present at sites of vascular inflammation. S100A9 is also expressed in epithelial cells constitutively or induced during dermatoses. S100A9 is a Calcium-binding protein. It has antimicrobial activity towards bacteria and fungi. It is important for resistance to invasion by pathogenic bacteria. It up-regulates transcription of genes that are under the control of NF-kappa-B. S100A9 plays a role in the development of endotoxic shock in response to bacterial lipopolysaccharide (LPS). It promotes tubulin polymerization when unphosphorylated. It also promotes phagocyte migration and infiltration of granulocytes at sites of wounding. It plays a role as a pro-inflammatory mediator in acute and chronic inflammation and up-regulates the release of IL8 and cell-surface expression of ICAM1.
